Hancock - one of Stoke City's most famous fans - is hoping to raise £100,000 for The Donna Louise Children's Hospice with Potters boss Pulis, who came up with the idea Kili 4 Kids after running last year's London Marathon.
Speaking to StaffsLive, local celebrity Hancock knows climbing Africa's highest mountain will be the toughest physical challenge of his life.
"Admittedly I haven't trained as well as I might have, but I've really stuck at it and my preparation hasn't gone badly.
"I've been going to the gym virtually every day and last month I did a practice climb up Snowdon, which was a very useful thing to do and actually quite enjoyable.
"My biggest worry is altitude sickness and we're having to ascend slowly to acclimatise to the conditions, so if all goes well it'll take us about six days to reach the peak."
Hancock is competing with Pulis to be the first to raise £50,000 in Team Hancock versus Team Pulis.
"Tony has the backing of thousands of Stoke supporters which is great, but we're both delighted with the support we've had already and people's capacity for generosity has been incredible," he said.
"It's a wonderful hospice and it needs to be open seven days-a-week which it can't afford at the moment, but now I'm really starting to believe we can raise £100,000 and that would be absolutely fantastic."
